脂肪酶结构与功能关系的FT-IR研究

摘    要:我们利用FTIR研究了脂肪酶在不同的冻干时间条件下二级结构的变化。研究了酶分子发挥摧化功能起重要作用的体系微环境对脂肪酶分子的二级结构的影响。结果表明,在最适催化温度范围内,酶分子的二级结构的确与其它温度下存在显著的不同。不同pH条件下,酶催化脂交换的活力相差较大,而酶分子的二级结构也确有明显的变化

Abstract:We investigated the structure of lipase using Fourier transform infrared. The results of FT IR spectra indicated that the dramatically changes in the secondary structure of lipase upon lyophilization of its aqueous solution had been occurred. The variation of secondary structure caused by different microenviroment had a good correlation with the changes of enzyme activity. The α helix and β sheet content lowered while the unordered structure increase in the range of the optical temperature. The effect of PH on secondary structure of lipase was mainly concerned with the structure change near the isoelectric point of enzyme.
