吐温-20敏化甲基蓝-蛋白质散射光谱研究与应用

摘    要:在非离子表面活性剂吐温-20存在下,甲基蓝与蛋白质作用形成离子缔合物,使共振瑞利散射(RRS)急剧增强.研究了相应的光谱特征、影响因素和适宜的反应条件.在pH=4.0和离子强度0.5 mmol5L-1的条件下, 甲基蓝与蛋白质作用在347 nm波长处产生特征RRS,吐温-20进一步敏化该RRS.不同蛋白质在一定浓度范围内与吐温-20敏化的散射强度呈线性关系,检出限在12.3~30.1 μg5L-1范围内.方法用于合成样品以及实际生物样品中蛋白质含量的测定, 结果满意.

Abstract:In the presence of non-ionic surfactant Tween-20,methyl blue(MB) could react with protein and form ion-association complex,leading to great enhancement of resonance rayleigh scattering(RRS) intensity.The corresponding characteristics of the spectra,the influencing factors and the optimum conditions of the reaction were studied.Under the conditions of pH=4.0 and ionic strength of 0.5 mmol5L-1,the interaction of MB with protein resulted in enhanced RRS signal at 347 nm.It was found that the interaction was s...
