甲壳素纯度的高效液相色谱法测定

摘    要:由于生物材料中甲壳素或壳聚糖是大分子,不溶于水,以及与蛋白质等结合在一起而难以定量测定,工业生产中尚无甲壳素或壳聚糖纯度的标准测定方法;采用甲壳素水解生成氨基葡萄糖,再用氯甲酸芴甲酯(FMOC—C1)进行衍生,利用液相色谱技术对衍生产物进行分析;结果表明,HPLC法是生物材料中甲壳素的一种灵敏的、特有的定性定量分析方法,为生物材料及工业化生产中甲壳素纯度的测定提供了一个方便的手段。

Abstract:Chitin is a kind of biological material with high relative molecular mass.Because of its unsolubility in water and combination with protein,it is difficult to determine its purity.A method for the determination of chitin purity is presented by high performance liquid chromatography.Chitin was hydrolyzed to glucosamine and then the products was derivatized using9_fluorenylmethoxycarbonyl chloride as a derivatization reagent.The results showed that the HPLC method offered a sensitive and specific tool for the quantification of chitin in biological material and industrial production.
