温泉瓶尔小草离体培养及种质试管保存培养基的筛选

摘    要:以温泉瓶尔小草新生幼叶为外植体,应用均匀设计法筛选其最适宜的离体培养和种质试管保存各阶段培养基.结果表明,最适合愈伤组织诱导的培养基为WPM+ZT0.50 mg·L-1+2,4-D1.90 mg·L-1,诱导率为96.5%;芽苗分化的培养基为WPM+ZT1.70 mg·L-1+NAA0.10 mg·L-1,分化率为99.0%;生根培养基为1/4MS+IAA0.05 mg·L-1+IBA0.03 mg·L-1,生根率达97.8%以上;试管保存培养基为WPM+KT1.10 mg·L-1+根皮苷3.20 mg·L-1,通过24个月的保存,芽苗生长率仅在6.5%以下.试验结果证明,成功建立了温泉瓶尔小草离体培养体系,常温条件下利用延缓生长的方法在试管内保存温泉瓶尔小草种质是可行的.

Abstract:The tender leaves of Ophioglossum thermale Kom.were used as explants,all suitable media of culture in vitro and germplasm conservation in vitro were screened through uniform design experiments.The results showed that WPM+ZT0.50 mg·L-1+2,4-D1.90 mg·L-1 was most suitable for callus induction with a induction rate of 96.5%;WPM+ZT1.70 mg·L-1 +NAA0.10 mg·L-1 was most suitable for shoots regeneration with a regeneration rate of 99.0%;1/4MS+IAA0.05 mg·L-1 +IBA0.03 mg·L-1 was most suitable for rooting with a rooting rate of 97.8%;WPM+KT1.10 mg·L-1+phloridzin3.20 mg·L-1was most suitable for in vitro germplasm preservation for 24 months with a growing rate of 6.5%.The results prove that In vitro culture system of Ophioglossum thermale has been successfully established,and the method of defering growth was practical for germplasm conservation in vitro at normal temperature.
