2种恩诺沙星注射液在鸡体内的药代动力学

摘    要:为比较考察由不同工艺制备的恩诺沙星注射液A和B在鸡体内的药动学过程,选用40日龄的健康海兰鸡进行了研究.A、B 2组分别肌注相应的恩诺沙星注射液(15 mg/kg),反相高效液相色谱法测定鸡血浆中恩诺沙星浓度,结果发现,A、B 2种注射液的恩诺沙星在血浆中的药动学行为均符合二室开放模型,主要动力学参数分别是:t1/2为9.68±0.02 h及18.60±0.05 h;t max为1.25±0.03 h及4.50±0.02 h;C max为1.88±0.12及1.40±0.20μg/mL;AUC(0-t)为19.72±0.21及26.76±4.15μg·mL-1·h.其中,注射液B的t1/2,t max及C max分别为注射液A的1.92,3.6和0.74倍,两者的药物动力学参数有显著性差异(P<0.05).结果表明,恩诺沙星注射液B肌注后吸收缓慢,药物的半衰期延长,临床应用72 h给药1次能维持对常见病原菌的有效血药浓度.

Abstract:In order to compare the pharmacokinetics of enrofloxacin injection made through different technologies, 15 mg/kg of injection A or B is injected into 40 - day - old healthy Hyling chickens. The reversed-phase high per- formance liquid chromatography method is adapted to measure the plasma concentration of enrofloxacin. Results show that the pharmacokinetic behavior of i. m. enrofloxacin injections A and B accord with the two-room open model. The key parameters are as follows: the t1/2 values for injections A and B are 9.68 ±0.02 h and 18.60 ±0.05 h, the Tmax values are 1.25 ±0.03 h and 4.50 ±0.02 h, the Cmax values are 1.88 ±0.12 and 1.40 ±0. 20 p.g/mL, and the AUC ( 0-t ) values are 19.72±0.21 and 26.76 ±4.15 μ g/mL. h, respectively. The t1/2, T1/2 and C values of the injection B are 1.92, 3.6 and 0.74 times higher than those of the injection A, all with significant differences ( P 〈 0.05 ). Aslo, it is shown that the drug absorption is slow and half-life period is prolonged for injection B compared with those of injection A. The effective drug concentration in blood can be main- tained in 72 h after one time clinical treatment.
