摘    要:在液氮内冻存的人外周血淋巴细胞,当冻存介质内不含低温保护剂时,细胞内呈现大量冰腔,细胞质、细胞核、细胞器和生物膜结构发生严重破坏.细胞的结冰是造成细胞死亡和功能丧失的初始原因.当冻存在含有10%二甲基亚砜或甘油的介质内,40天时,绝大部分淋巴细胞具有完好的超微结构,少量细胞遭受冷冻损伤,主要表现在细胞器.如线粒体膨胀、(?)结构破坏等.冻存一年时,部分细胞的细胞核形状和结构也发生了深刻的变化,如细胞核分叶,核膜呈锯齿状皱缩,染色质分布不匀等.

Abstract:The human peripheral lymphocytes cryopreserved in liquid nitrogen without cryoprotectant in the freezing media demonstrated a serious aberration in the ultrastructure of the freezing cells. A vast number of small ice cavities were observed and the cytoplasm, nucleus, cell organelles and biomembrane were dissolved in almost all cells. Ice formatiop seemed to be the main cause of the functional loss and death of these lymphocytes. The lymphocytes which were cryopreserved in media containing 10%Me2SO or glycerol for 40 days displayed an intact ultrastructure in most of the preserved cells. Only a few cells suffered freezing damage after cryopreser-vation in which the orgenelles were deformed with swelling mitochondria and broken crest.After one year's cryopreservation, intensive changes were found in part of the freezing cells, such as the polymorphic nuclers, shrinkage of nuclear membrane and the aggregated chromatin.Changes of the ultrastructure in preserved lymphocytes were likely to to be related with the length of the freezing period.
