偶联亲水层的牛血清白蛋白手性固定相的制备

摘    要:在保证完全拆分的前提下,为了减少拆分所需的时间和流动相,对常用的羰基二咪唑活化制备蛋白质手性固定相的方法进行改进。在偶联牛血清白蛋白之前先偶联一层亲水层,降低溶质与固定相的疏水作用从而减小了被拆分对映体的保留时间(保留时间由原来的39min减小至8.74min)。分别考察了各种流动相条件和进样条件对色氨酸对映体保留因子和分离因子的影响。

Abstract:Under condition of total enantiomeric separation, in order to decrease needed time and mobile phase, normal method to prepare chiral stationary phase with 1,1-carbonyldiimidazole was improved. A film of hydrophilic chitosan was covalently coupled on the aminopropyl silica before immobilization of bovine serum albumin . It can weaken hydrophobic interaction between enantiomers and albumin and decrease the retention time of the enantiomers and the revention the decreased from 39 min to 8.74 min. And the effects of varied conditions of mobile phase and the sample on the retetion factor and separation factor of D,L-tryptophan were investigated.
