摘    要:建立了HPLC法测定鸡腿菇水解液中单糖、低聚糖及多糖含量的分析方法.选用Aminex HPX-87H柱,示差折光检测器,柱温50 ℃,5 mmol/L的硫酸为流动相,流速0.4 mL/min,进样量20 μL,外标法定量.结果表明,单糖、低聚糖及多糖均得到较好分离,其峰面积与质量浓度线性关系良好,相关系数r~2值为0.991 2 ~0.999 7,回收率为98% ~106%.采用HPLC-示差法与气相色谱法测定多糖、低聚糖及单糖相比,样品无需衍生处理,过滤后可直接进样检测.该法可同时测定原料水解液中可能存在的主要单糖、低聚糖及多糖.

Abstract:A high performance liquid chromatography(HPLC) method was established for determination of monosaccharides,oligosaccharide and polysaccharide in the liquors of hydrolysis on Coprinus comatus.The sample was measured by Aminex HPX-87H column(300 mm×7.8 mm) with differential refractive index detector at 50 ℃.The mobile phase was 5 mmol/L H_2SO_4 with flow rate of 0.4 mL/min and the injection volume was 20 μL.The result indicated that mixture of monosaccharides,oligosaccharide and polysaccharide was successfully separated and a good relationship was obtained between peak area and concentration of monosaccharides,oligosaccharide and polysaccharide.The correlation coefficients of those matters ranged from 0.991 2 to 0.999 7 and the corresponding average recoveries were in the rang of 98%-106%.Compared with gas chromatographic method,the method could directly detect monosaccharides,oligosaccharide and polysaccharide samples after filtration without derivative processing.The novel HPLC method for the simultaneous quantification of monosaccharides,oligosaccharide and polysaccharide is simple,rapid and accurate.
