Abstract:To establish a rapid,simple,sensitive enzymatic end point assay of MPT-NAG using metal ions as stabilizer,the analysis is based on the fact that the MPT-NAG,when hydrolyzed in the presence of N-acetyl-beta-D-glucosaminidase,liberates MPT as the chromogen.The absorbence of the chromogen is direct ratio to the concentration of MPT-NAG.The result shows that the method is liner in the range of 0~50 mmol/L,the average rate of recovery is 101.5%,the within run and between run CV are 0.010 5~0.026 4 and 0.013 2~0.031 6,respectively.Comparing this method with HPLC,as the regression equation: y=1.021 0 x + 0.882 1,r=0.998 9.This method was simple,sensitive and suitable for application.
