摘    要:以乙醇钾为脱氢试剂,吖啶酮为原料合成了10-甲基-吖啶酮,将其硝基化后成功合成了一种新型吖啶酮衍生物—10-甲基-3-硝基-吖啶酮(MNA)。采用循环伏安法对MNA及其与蛋白质相互作用的电化学行为进行研究,并建立了差示脉冲伏安法测定蛋白质的新方法。在PBS缓冲溶液(pH7.0)中,MNA与蛋白质相互作用形成超分子复合物,使其氧化还原峰电流降低,降低值同所加入的牛血清白蛋白(BSA)和人血清白蛋白(HSA)分别在0.4~14 mg/L和0.04~0.28 mg/L范围内呈良好的线性关系,并已应用于实际样品的测定。

Abstract:Potassium ethoxide was used as dehydrogenation reagent,and 10-methyl-acridone was synthesized from acridone.Then,a kind of new acridone derivative-10-methyl-3-nitro-acridone(MNA) was synthesized by nitration of 10-methyl-acridone.The electrochemical behavior of the interaction of MNA with proteinum was investigated by cyclic voltammetry and a differential pulse voltammetric method has been proposed for the determination of the protein with MNA.In pH 7.0 phosphate buffer solutions(PBS),an electrochemically non-active complex was formed via intercalation of MNA with protein.On addition of protein into the MNA solution,the oxidative peak current of MNA decreases.The decrease of oxidative peak current of MNA was proportional to BSA and HSA concentration in the range of 0.4-14 mg/L and 0.04-0.28 mg/L,respectively.This method had been applied to the analysis of HSA samples.
