多功能蛋白增活器的研制及性能研究

摘    要:自制了一种新型的多功能蛋白增活器,它具有同时除变性剂、分离杂蛋白、复性目标蛋白及便于回收变性剂的功能。考查了其对蛋白的分离、复性性能。发现这种多功能蛋白增活器可对6种标准蛋白进行良好地分离。同时对盐酸胍变性的溶菌酶和核糖核酸酶完全复性。通过测定压力-流速曲线,发现多功能蛋白增活器的压力均远远低于普通色谱柱。将其应用于基因工程发酵的粒细胞集落因子(G-CSF)的分离纯化,可使纯度接近于100%,生物活性大于常规方法3倍

Abstract:A multi functional increment activator for protein (MIAP) has been invented. The MIAP has four functions simultaneously: removing denaturing agent, separation from impure proteins, renaturation of aim protein, and recycling the denaturing agent. The efficiencies of the MIAP were also investigated. such as the separation efficiency, renaturation efficiency and back pressure. It indicates that six kinds of proteins could be separated completely by using the MIAP. In addition, the denatured lysozyme and ribonulease in guanidine hydrochloride could also be renatured by various kinds of MIAP. From the plot of back pressure vs flow rate, the back pressure of the MIAP is much lower than the usual chromatographic column. During the preparation of recombinant human granulocyte colony stimulating factor (rhG CSF) by the MIAP, nearly 100% purity could be achieved and its bioactivity recovery is far more than 3 times.
