Abstract: | A high performance liquid chromatography method is presented for the determination of valproic acid levels in human plasma. The method was based on pre-column derivatization using N-(1-naphthyl)ethylenediamine as a new labeling agent. The calibration curve was linear in the investigated concentration range between 0.1 and 100 μg mL?1 and showed good accuracy and reproducibility. The assay provided a limit of quantification of 0.1 μg mL?1 for valproic acid and a limit of detection of 10 ng mL?1, respectively. The presented method was successfully applied to the determination of valproic acid levels in plasma after oral administration of 600 or 800 mg of sodium valproate. |
