浙江沿海大弹涂鱼、弹涂鱼的线粒体COI基因序列差异及其系统发育

摘    要:为探讨浙江沿海大弹涂鱼和弹涂鱼线粒体细胞色素氧化酶I号(COI)基因序列差异及其系统发育, 对两者进行了COI扩增、测序和分析. 结果显示: 扩增得到大弹涂鱼、弹涂鱼COI基因序列长度分别为1554bp和1556bp; 大弹涂鱼和弹涂鱼COI基因序列中, 所占比例(分别为55.8%和56.0%)均高于 所占比例(分别为44.2%和44.0%); 大弹涂鱼和弹涂鱼的种内平均遗传距离分别为0.002和0.003, 两者的种间遗传距离为0.183. 构建的系统发育树显示, 大弹涂鱼与红狼牙虾虎鱼、孔虾虎鱼、犬齿背眼虾虎鱼聚为一簇(大弹涂鱼COI序列与三者的同源性分别为91.0%、92.0%和88.0%), 弹涂鱼与青弹涂鱼聚为一簇(两者COI序列同源性为99.0%)后与大弹涂鱼所在的簇相聚. 研究结果可为探讨大弹涂鱼、弹涂鱼的系统发育及建立基于COI序列的弹涂鱼类鉴定方法提供参考资料.

Abstract:In order to explore the variability of mitochondrial cytochrome oxidase I (COI) gene and phylogeny of Boleophthalmus (Boleophthalmus pectinirostris) as well as Mudskippers (Periophthalmus cantonensis) in coastal of Zhejiang Province, we sequence and analyze COI gene of these two fish species. The results show: the length of the COI gene of B. pectinirostris and P. cantonensis is 1554bp and 1556bp, respectively. In the B. pectinirostris and P. cantonensis COI sequences, the content of (55.8% and 56.0%, respectively) is higher than (44.2% and 44.0%, respectively). The average intra-specific genetic distance of B. pectinirostris and P. cantonensis is respectively identified as being 0.002 and 0.003. The genetic distance between the two species is found to be 0.183. The phylogenetic tree indicates that B. pectinirostris with Odontamblyopus rubicundus, Trypauchen vagina, Oxuderces dentatus are grouped into a cluster (The homology between B. pectinirostris and the other three kind of fishes is 91.0%, 92.2% and 88.0%, respectively); the P. cantonensis and Scartelaos histophorus gather into a cluster (the sequence homology is 99%) together with the cluster of B. pectinirostris. The results are expected to provide the reference for exploring the phylogenetic of B. pectinirostris and P. cantonensis, as well as identification method of Mudskippers based on COI
