纤维与水溶液状态下DNA空间构象的激光拉曼光谱分析

摘    要:本文用改进的Brown比值、DNA分子拉曼强度与试样环境相对湿度的函数关系两种方法,分析了反映小牛胸腺DNA在纤维状态与水溶液中构象的激光拉曼数据。我们认为,应用1807/I1100计算DNA纤维中A型构象的比例;在水溶液中,DNA以B型构象为主,但还存在A型构象,这与紫外共振激光拉曼分析结果相同,对此进行了有关机制的讨论。

Laser Raman Spectroscopic Analyses of DNA Spatial Conformation in Flber and Aqueous Solution States
Abstract:By the improved Brown’s ratio method and the functional relationbetweeii the DNA molecular Raman intensitv and the relative humidity of testenvironn1cnt, the Laser Raman data of spatial conformation of calf thymusDNA in fiber and aqueous solution states have been analysed. The experimen-tal results evidencedthat the 1807/11 100 ratio should have been used for thecalculation of type一Aform content in the DNA fiber, The type一Bform isdominant in the DNA aqueous solution,but the type一Aform exist also in,similar to the results of Ultraviolet Resonance Raman Spectroscopie analyses。In view of above,the concerned mechanism was discussed in this paper。
