药根碱与溶菌酶相互作用的光谱研究

摘    要:利用荧光光谱和红外光谱研究了药根碱与溶菌酶的结合反应。通过Stern-Volmer's方程分别计算了不同温度下反应的结合常数。FTIR光谱结果表明:药根碱与溶菌酶键合改变了溶菌酶原有的构象,使溶菌酶的α-螺旋和β-折叠的含量发生变化。根据Foerster非辐射能量转移理论,计算出药根碱与溶菌酶之间的结合距离为5.39nm。

Abstract:Optical properties of the jatrorrhizine-lysozyme complex were characterized by fluorescence and Fourier transform infrared(FT-IR)spectra.The binding constants at different temperature were calculated according to Stern-Volmer's equation.The protein conformation is altered with enhancement of α-helices and decrease of β-turn structure.The distance from the tryptophan residue(donor)to the bound drug(acceptor)in lysozyme was calculated as 5.39nm through Foerster non-radiation energy transform theory.
