TTC-脱氢酶活性测定中标准曲线的影响因素研究

摘    要:此文采用722S型可见分光光度计,以连二亚硫酸钠为还原剂,对传统TTC-脱氢酶活性测定法中影响标准曲线制作的因素进行了研究,确定了最佳测定条件为:最大吸收峰在475nm处;连二亚硫酸钠的最佳投量为5.0mg/5mL;11℃溶液的保存及整个实验过程应避光。最佳反应时间为30min。

Abstract:In this paper the factors which influence the standard curve of determination are investigated by using 722S type visible pectrophotometer and sodium hydrosulfite as reductant in the measure method conventional TIC - Dehydrogenase activity. The optimum conditions are as follows: the maximum absorption peak is at the point of 475 nm; the best dosage of sodium dithionite is 5. 0 mg/5mL ; preservation of TTC solution and the whole test process should be in darkness; the optimum reaction time is 30 rain.
