乙基紫标记分光光度法测定脱氧核糖核酸

摘    要:研究了阳离子染料乙基紫与脱氧核糖核酸的结合反应,在ph=6.4-7.4的条件下,乙基紫在595nm处有最大吸收峰,当加入DNA后,乙基紫的吸收峰显著下降,而在507nm处出现新的吸收峰。以乙基紫为标记物,根据其595nm吸收峰下降的程度,可用于DNA的定量测定。

Abstract:The binding reaction of DNA (calf thymus deoxyribonucleic acid) with dye ethyl violet has been studied. In the medium of pH range 6 .4 - 7. 4, ethyl violet showed a decrease in absorption at the wavelength of its maximal absorption peak (595 um) on binding to DNA, and at the same time a new peak of the binding product appeared at 507 um. A novel method for the determination of DNA at 595 um using ethyl violet as a labeling agent was developed. The method is of high sensitivity as well as selectivity, and has been used to determine DNA in synthetic Samples with satisfactory results.
