雄黄对大鼠脑组织中单胺类神经递质及其代谢产物的影响

摘    要:将32只Wistar大鼠随机分为对照组(口服0.5%羧甲基纤维素钠溶液)以及低剂量(0.3g/kg)、中剂量(0.9g/kg)和高剂量(2.7g/kg)雄黄混悬液处理组,通过4周连续灌胃给服雄黄混悬液;采用高效液相色谱-电化学法测定了大鼠脑组织中单胺类神经递质及其代谢产物的含量,研究了雄黄对大鼠脑组织单胺类神经递质及其代谢产物的影响.结果表明,与对照组比较,雄黄染毒组大鼠脑组织中NE、DOPAC和DA含量均呈增加趋势,而5-HIAA呈降低趋势.雄黄可对大鼠脑组织单胺类神经递质及其代谢产物产生影响,单胺类神经能系统可能是雄黄毒性作用的靶点之一.

Abstract:32 Wistar rats were categorized into control group and low dosage, moderate dosage, and high dosage realgar-treated groups. The control group was orally fed with the solution of 0.5 % sodium carboxymethylcellulose for 4 weeks, while the realgar-treated groups were treated with realgar suspension by gastric perfusion at a dosage of 0.3 g/kg, 0.9 g/kg, and 2.7 g/kg, respectively, for the same duration. The content of monoamine neurotransmitters and their metabolites was determined by means of high performance liquid chromatography equipped with an electrochemical detector, and the effect of realgar on monoamine neurotransmitters and their metabolites in the brain tissues of the rats was investigated. Result shows that the contents of NE, DOPAC and DA in.the brain tissues of realgar-treated groups tend to increase while the content of 5-HIAA tends to decrease as compared with the control group. In one word, realgar can affect monoamine neurotransmitters and their metabolites, and monoaminergic system may be one of the toxic targets of realgar.
