傅里叶变换红外光谱研究鲤鱼雌激素受体与邻苯二甲酸酯的相互作用

摘    要:通过电泳结合电洗脱(SDS-PAGE)的方法分离纯化鲤鱼体内的雌激素受体,得到高纯度的受体;再通过傅里叶变换红外光谱探讨雌激素受体与邻苯二甲酸二正丁酯(DBP)结合后其结构的变化,通过实验发现,配体DBP能与雌激素受体结合并对雌激素受体的结构产生了明显的影响。

Abstract:Phthalic acid esters(PAEs) were used as plasticizers in plastic industry widely,which is one kind of environmental endocrine disruptors.They had a longer and more permanent effect on animals,they can disturb normal physiologic processes which were mediated by the estrogen receptors.The method of sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE) and electroelution to purify the estrogen receptor from carp,and the estrogen receptor of high purify was obtained.Fourier transform infrared spectroscopy was used to study the structure of estrogen receptor change that interact with DBP.The result prove that ligand DBP can bind with the estrogen receptor,and have an obvious effect on the structure of estrogen receptor.
