摘    要:采用超声波法提取关黄柏中有效成分盐酸小檗碱,应用正交试验法筛选盐酸小檗碱最佳提取工艺。采用高分离度快速液相色谱(RRLC)测定关黄柏中有效成分盐酸小檗碱的含量。色谱条件为:色谱柱:XDB-C18(4.6mm×50mm,1.8μm);流动相:乙腈-0.3%磷酸缓冲溶液(29∶71);检测波长:266nm;流速:0.7mL/min;柱温:25℃;进样量:2μL。盐酸小檗碱在0.075—0.3μg范围内线性关系良好,r=0.9998,其平均回收率为99.2%,RSD为1.48%。本法操作快速简便、准确可靠,适用于关黄柏中盐酸小檗碱的含量测定。

Abstract:The active principle berberine hydrochloride in Phellodendron amurense Rupr was extracted by ultrasonic wave method,and the optimal extraction process of berberine hydrochlorid was choosed by orthogonal test.The content of the active principle berberine hydrochlorid in Phellodendron amurense Rupr was determined by resolution rapid liquid chromatogram(RRLC).The chromatogramic conditions were as follows:column of Agilent XDB C18(4.6mm×50mm,1.8μm),the mobile phase consists of acetonitrile-0.3% phosphoric acid(29∶71),the detection wavelength of 266nm,the flow rate of 0.7mL/min,the column temperature of 25℃ and the sample size of 2μL.There was a good linear relationship for berberine hydrochlorid in the range of 0.075—0.3μg with correlation coefficient of 0.9998,and the average recovery was 99.2% with RSD of 1.48%.The method is fast,simple,accurate and reliable,and is suitable to determine the content of correlation coefficient in Phellodendron amurense Rupr.
