Abstract:n this paper, some factors affecting the purification of interleukin-2 (IL-2) have been summarized, such as ion strength, gradient program and selection of flow rate etc. We can conclude that in reversed-phase high performance liquid chromatography, for sample in high salt concentration, the salt should be removed before the sample was injected into the column in order to avoid sedimentation in the column and pipeline. We also find that very small changes of elution gradient will strongly affect the results of separation. With different samples, we should find out the most suitable gradient program by repeated experiments. Too-high or too-low flow rate is unfavourable to the results of separation. In order to avoid air bubbles and equilibrate the column, while stopping gradient program, the method that concentration of organic solvent is cut down in reverse gradient program can be used.
