大豆聚丙烯酰胺凝胶电泳制胶浓度选择

摘    要:本文提出在聚丙烯酰胺凝胶电泳中,对未知样品宜先在C%=4.8%,T%=2~5%或C%=2.6% T%=5~10%的系列浓度分离胶及C%=20%,T%=3.2%的浓缩胶中作圆盘电泳,选择该样品的最适分离胶浓度。然后再按此浓度作垂直平板电泳,并作加样量梯度实验,找出加样最适量,以达到样品在聚丙烯酰胺凝胶垂直平板电泳中的最佳分辨率。

Abstract:It is put foreward in this paper that Diac-PAGE should be made first for soybeans samples in a series of C%=4.8% T%=2~5% or C%=2.6% T%=5~10% lower sparating gel (or running gel)and C%=20% T%=3.2% upper gel (stacking gel). Then the optimum lower searating gel concentration sel should be ected. In order to attain the best identification of the sample in vertical PAGE the theaptimum concentration should be used and the quantitive degree of sample should be made for determining the optimum quantity.
