| 摘 要： | 本实验室从正常狗脑中又分离获得一个暂称这为“６０ａ”的肽。冻干粉经ＳｅｐｈａｄｅｒＧ－５０和羧甲基纤维素ＣＭ２２柱层析，再经反相高效液相层析纯化得最后样品，经ＳＤＳ电泳和等电聚焦电泳鉴定均为一条带，分子量约８０００，等电点在ｐＨ７．８左右；Ｎ－末端氨基酸为Ｍｅｔ；自Ｎ－末端的部分氨基酸序列为：Ｈ２Ｎ－Ｎｅｔ－Ｌｅｕ－Ｓｅｒ－Ｐｒｏ－Ａｌｓ－Ａｓｐ－Ｌｅｕ－Ｔｈｒ－Ａｓｐ－Ｉｌｅ－Ｌｅｕ－Ｐｈｅ－Ｔ

| Abstract: | A pure peptide named 60a from dog brain was obtained. The whole dog brain was homogenized and extracted with 0.1mol/L acetic acid. The supernatant was lyophilized and the residue was dissolved in 0.1mol/L acetic acid and was allowed to go through Sephadex G-50 column (twice), CM22 cellulose. The sample obtained was applied to a rp-HPLC with C18 column. The sample was named 60a peptide, and showed only one band on SDS-polyacrylamide gel electrophoresis or isoelectric focusing. Its isoelectric point was around pH 7.8 and M. W. was about 8000 dalton. The sequince of the N-terminal 46 amino acid residues has determined. Its N-terminal amino acid was Met. The 60a peptide showed a significant anti-morphine activity when intravetricularly injected to the mice. |
