羽衣甘蓝组织培养研究

摘    要:以羽衣甘蓝的花蕾、花茎和腋芽为外植体,进行组织培养试验.结果表明:花蕾、花茎消毒时间以5~7min为好,腋芽消毒时间以8 min为宜,以腋芽为外植体,以MS基础培养 6-苄基腺嘌呤(BA)2 mg 萘乙酸(NAA)0.2 mg 3%蔗糖进行培养较好,继代培养以MS基础培养 BA 3 mg NAA 0.2 mg 3%蔗糖增殖率最高;在生根阶段,以50%MS(1 L) NAA0.4 mg 1%蔗糖生根效果最好,生根率达94.79%;移栽基质以蛭石最好,羽衣甘蓝组培苗成活率达86.7%,且生长健壮.

Abstract:The flower buds,pedicels and axillary of ornamental kale were cultured in vitro.The optimum disinfection time was 5~7 minutes for flower buds and pedicels,and was 8 minutes for axillary.MS 2 mg/L BA 0.2 mg/L NAA 3% cane sugar was optimum medium for induction of axillary,MS 3 mg/L BA 0.2 mg/L NAA 3% cane sugar was the best medium for its proliferation,and 50%MS 0.4 mg/L NAA 1% cane sugar was the best medium for its rooting and the rooting rate was 94.8%.Vermiculite was the optimum matrix for its transplant,and the survival rate was 86.7%.
