小鼠血浆中染料木素水溶性代谢产物和染料木素脂肪酸酯的分离及测定

摘    要:本文建立了小鼠灌胃染料木素单体后血浆中水溶性染料木素代谢产物和染料木素脂肪酸酯的分离及测定方法。血浆样品经乙酸乙酯萃取后上Sephadex LH-20柱,分别用体积比1∶1的正己烷/氯仿和甲醇洗脱,染料木素脂肪酸酯用脂肪酶酶解后转化成染料木素,水溶性代谢产物用葡萄糖醛酸酶及硫酸酯酶水解成染料木素,然后采用液相色谱串联飞行时间质谱(Q-TOF LC/MS)检测染料木素。血浆中染料木素在10~10000 ng/mL范围内线性关系良好,检测限为1 ng/mL。10批次小鼠血浆中水溶性代谢产物平均为526.006 ng/mL,染料木素脂肪酸酯平均为58.976 ng/mL。采用SephadexLH-20柱具有良好的分离效果,脂肪酶水解染料木素脂肪酸酯稳定、专一性强,用Q-TOF LC/MS检测染料木素快速、灵敏。

Abstract:To separate and determine the water-soluble metabolites of genistein and its fatty acid ester in mice’s plasma which were exposed to genistein by gavage,the plasma samples were eluted by n-hexane/chloroform(1∶1 V/V) and methanol successively through Sephadex LH-20 after being extracted by ethyl acetate.Genstein fatty acid ester was changed into genstein via lipase’ enzymolysis,then the samples were determined using Q-TOF LC/MS.The linear range for genistein was 10~10 000 ng/mL.The limit of detection was 1.0 ng/mL.The average water-soluble metabolites were 526.001 ng/mL and genistein fatty acid ester was 58.977 ng/mL in 10 batches mouse plasma.
