摘    要:利用一种新的高效液相色谱洗脱技术,建立了 pH缓冲液/离子对二元洗脱模式,对双峰驼精清的两种提取物的反相柱上进行了分离,得到了较纯的活性蛋白组分。该法操作简单、快速、比常规HPLC有更高的选择性及峰容量,并可减少活性蛋白的失活。

Abstract:A new HPLC elution technique consisting of pH/ion-pair bimodal elution was es- tablished to separate and purify the active protein(s) from camelus seminal plasma. ln this method the inactive components were first eluted by the pH buffer and the active components were then eluted by the ion-pair mobile phase. The influences of pH and switching time in the bimodal elution on the retention of the active protein were investigated. The optimum chromatographic condition was given. The pure active protein(s) was obtained. This method is simple and quick, with higher selectivity and peak capacity than conventional reversed phase chromatogrphy. It can also reduce the risk of denaturation to the active protein.
