摘    要:长链非编码RNA(long noncoding RNA,lncRNA)是一类长度超过200 nt,并不编码蛋白质的RNA分子。近年来关于lncRNA的研究逐渐成为热点,但绝大部分lncRNA的功能仍不清楚。本实验室前期Arraystar lncRNA芯片分析发现lncRNA21905在有丝分裂期的表达量显著高于细胞分裂间期,提示该lncRNA在有丝分裂期中可能有一定功能。研究通过siRNA干扰技术,在肿瘤细胞中敲低lncRNA21905,发现敲低该lncRNA可导致肿瘤细胞有丝分裂期的明显阻滞。此外,TANRIC数据库分析表明,lncRNA21905高表达的肾透明细胞癌患者预后较差。综上所述,研究表明lncRNA21905在肿瘤细胞有丝分裂期中发挥了重要调控作用,这可能为进一步揭示其参与肾透明细胞癌预后提供理论基础。

Abstract:Long noncoding RNA (lncRNA) is a kind of noncoding RNA with length more than 200 nucleotides. The researches on lncRNA have increased rapidly, but the functions of most lncRNA are still unknown. With the help of Arraystar lncRNA chip analysis, we found that the expression of lncRNA21905 is significantly higher in mitosis than that in interphase, which indicate that lncRNA21905 may have function in mitosis. In this research, we depleted lncRNA21905 by siRNA, and found that knockdown of lncRNA21905 can induce obvious mitotic arrest in tumor cells. In addition, through the TANRIC database analysis, we also found that the prognosis of renal clear cell carcinoma (RCCC) patients with high expression of lncRNA21905 is poor. Above all, this research indicate that lncRNA21905 have an important function on the regulation of mitosis in tumor cells, which may provide a theoretical basis for further revealing its involvement in the prognosis of RCCC.
