酰胺型手性固定相反相拆分布洛芬药物对映体

摘    要:利用酰胺型手性固定相反相拆分了布洛芬对映异构体,用有0.01mol/L NH4Ac的甲醇和水作流动相。在优化分离条件的同时,研究了不同的有机调节和对分离的影响,并探讨了分离的机理。

Abstract:By using reversed-phase high performance liquid chromatography with amide derivative [ (R)-1-naphthylglycine & 3,5-dinitrobenzoic acid] as chiral stationary phase (CSP) , the direct resolution of laconic ibuprofen was investigated . "Die resolution was optimized by varying the concentration of methanol, and the pH in the mobile phase. The organic modifier was also altered by substituting acetonitrile and tetrahydrofuran (THF) for methanol. The optimized mobile phase was methanol/H2O (70/30, V/V ), containing 0.01 mol/L NH4Ac and pH 5. 9 (with RAc). Although the racemic ibuprofen couldn't been direct resolution by HPLC with urea derivative [( S)-Valine& ( R)-l-( a-naphthyl)ethylamine] as the CSP , the resolutions of ibuprofen with these two CSPs are agreement with the three-point rule.
