脱落酸对伊文思蓝的荧光猝灭机理研究及其含量测定

摘    要:利用荧光光谱研究了脱落酸(Abscisic acid)与伊文思蓝(Evans blue)的相互作用。用266 nm激发光激发时,伊文思蓝的最大发射波长为396 nm。伊文思蓝与脱落酸相互作用,使伊文思蓝发生荧光猝灭,根据Stern-Volmer方程研究了荧光猝灭的类型及机理,实验证明脱落酸与伊文思蓝发生的静态猝灭,即脱落酸和伊文思蓝形成了一种稳定的复合物。伊文思蓝的相对荧光强度与脱落酸的浓度线性相关,线性方程为F0/F=0.78153+3.9153×104c,线性相关系数为0.9976。通过实验求算了脱落酸与伊文思蓝的结合点常数KABA-EB=1.838×105L/mol和结合点数n=1.171,由此建立了测定植物中脱落酸的新方法。

Abstract:The interaction between evans blue (EB) and abscisic acid (ABA) was studied by the fluorescence spectroscopy. The maximum emission wavelength was 396 nm as the excited wavelength was 266 nm. The relative fluorescence intensity of evans blue (EB) decreased in the presence of abscisic acid (ABA), which showed that fluorescence quenching occurred. A linear relationship was found between the relative fluorescence intensity of EB and the concentration of ABA. The linear equation was Fo/F =0. 78153 + 3. 915 3 ×10^4c with linear correlation coefficient of 0. 997 6. The mechanism of the fluorescence quenching was discussed. The binding constant and the number of binding sites were 1. 838 ×10^5 L/mol and 1. 171 respectively. A new method for determination of EB was established.
