叶酸与氨基酸相互作用的荧光光谱

摘    要:在模拟动物生理条件下,利用荧光光谱法在分子水平上研究了叶酸同色氨酸、酪氨酸、苯丙氨酸的相互作用;根据双对数方程求出了不同温度下两者反应的结合常数和结合位点数.结果表明:叶酸能导致氨基酸的荧光猝灭,三种氨基酸的猝灭机制均为静态猝灭;从相应的热力学参数可知,色氨酸和酪氨酸与叶酸之间的主要作用力为疏水作用力,而苯丙氨酸与叶酸之间的主要作用力为氢键和范德华力.

Abstract:The interactions between folie acid and amino acids including thetryptophan, tyro- sine, phenylalanine were investigated by fluorescence spectroscopy under simulated physiologi- cal condition of animal body and the binding constants and the number of binding sites were calculated with double logarithmic equation. Results show that folie acid is able to quench the fluorescence intensity of the amino acids and the quenching of the three amino acids by folie acid is a static quenching process. Relevant thermodynamic parameters demonstrate that hydro- phobic force plays a major role in the binding of folie acid with tryptophan and tyrosine, while both hydrogen bond and Van der Waals force play a major role in the binding of folie acid with phenylalanine.
