Use of NOESY for estimation of coupling constants in the DNA backbone

Abstract:We demonstrate here the use of the two-dimensional NOESY technique for measurement of the most elusive coupling constants, J(H4′-H5′), J(H4′-H5″), J(H3′-H4′) and J(H3′-P) in long DNA segments. The band selective BURP family of pulses has been used in the NOESY pulse sequence for achieving high resolution in the spectra and the coupling constants have been estimated by simulating cross-sections through H1′-H4′ and H8-H3′ cross-peaks in two illustrative cases.
