5-甲基色胺酰胺化反应的研究

摘    要:设计了色胺酰胺化的一锅法合成方法. 由色胺盐酸盐现场与碱中和释放出的游离色胺, 与羧酸的活化酯反应, 得到酰胺. 考察了不同的碱、色胺盐酸盐与碱的比率及反应时间对酰胺化反应产率的影响. 结果表明: 在添加三乙胺或氢氧化钠等碱的条件下, 通过一锅法反应, 可以提高合成目标化合物的产率. 该合成方法操作简单、反应条件温和、反应时间短, 产率达90%以上.

Abstract:One-pot synthetic method for the amidation of 5-methyltryptamine hydrochloride is proposed. The free 5-methyltryptamine, which is released by neutralization of 5-methyltryptamine hydrochloride with a base in situ, reacts with the activated ester to yield amide. The influences of the bases, the ratio of amine hydrochloride and base, and the reaction time on the amidation are systematically investigated. The results show that in the presence of triethylamine or sodium hydroxide, amide can be obtained in moderate to high yields. One-pot procedure is fast-paced and convenient, and the optimized yields can reach as high as 90%
