水稻花药特异表达基因启动子的扩增及克隆

摘    要:采用DNA聚合酶链式反应(PCR)扩增技术,以水稻黄化苗总DNA为模板成功地扩增到水稻花药特异表达基因启动子Osg6B的770bp和960bp两个片段Osg6Ba和Osg6Bb.并将其克隆到pUC18上形成完整的Osg6B,这为通过基因工程方法进行水稻杂种优势利用奠定了基础.

Abstract:Calli of four wild rices were desiccated in a petri dish with a single layer of filter paper prior to transfer to the regeneration medium.The plant regeneration frequencies of three wild rices were increased at different degrees.Desiccation of calli resulted in the signifcant declining of indole 3 acetic acid(IAA) and water soluble protein (WSP)content,but activity of peroxidase iszyme was increased. With desiccation,the increasing of activity of peroxidase iszyme might result in the decreasing of IAA content,and improved plant regeneration frequency of calli of wild rice.
