粒/巨噬细胞集落因子在中耳胆脂瘤中的表达和作用研究

摘    要:为探讨粒/巨噬细胞集落因子(GM-CSF)在继发性中耳胆脂瘤中的表达情况,分析其在胆脂瘤上皮增殖、骨质破坏特性中的作用,应用免疫组化SABC染色方法和计算机图像分析法,检测25例胆脂意上皮和10例正常外耳道皮肤中GM-CSF的表达情况。结果表明,GM-CSF在22例胆脂瘤上皮组织的全层细胞的胞浆中有强表达,阳性率为88%;GM-CSP在10例正常外耳道皮肤上皮中有弱表达,阳性率为40%。染色结果经计算机图像分析仪定量分析,两者的平均光密度分别为2.332和1.474,差异呈高度显著性(P<0.05)。说明中耳胆脂瘤上皮中GM-CSF的含量显著高于正常外耳边皮肤的含量。推测GM-CSF可能参与了胆脂瘤的发生、发展及可能参与了胆脂病上皮增殖、骨质破坏的作用。

Abstract:The objective of the research is to evaluate the role of GM-CSF in acquired middle earcholesteatoma. For this,the specimens from the acquired middle ear cholesteatoma tissue of 25 casesand the nomal external ear skin of 10 cases were examined by immunohistochemical SABC method andcomputer image analysis. In 22 cholesteatoma cases,GM-CSF expresses in the cytoplasm of the full ep-ithelial tissue cells,the percentage of positive is 88%. Whereas in 10 cases of normal external ear skin,the percentage of positive is 40%. The results of the computer image quantitative analysis systemshow that the mean optical density of GM-CSF in the two types of tissue were 2. 322 and 1. 464 re-spectively with a significant difference between them(P<0. 05). The expression level of GM-CSF ofcholesteatoma epithelial tissue is higher than that of normal external auditory meatal skin epithelialtissue. This study suggests that GM-CSF is involved in the development and destructive effects of ac-quired middle ear cholesteatoma.
