从水稻悬浮细胞获得原生质体再生植株

摘    要:以长香稻(Oryza sativa L.)(糯稻)的幼嫩种子限为材料,在MS和N6基本培养基上诱导产生结构松散的愈伤组织,在AA培养基中进行液体振荡培养,悬浮细胞经酶解后获得了少在性较好的原生质体,试验证明,水稻悬浮细胞是分离原生质体较理想的材料,在附加2.4-Dlmg/L(以下单位同),KT0.2,谷氨酰胺867,天门冬胺酸266的MS琼脂糖培养基中,诱导出了大量愈伤组织;在含KT2,NAA0.

Abstract:Loose ealli from tender roots of oryza sativa L. on MS and N_6 media are the better materinl for producing protoplasts. More protoplasts were isolated from callus-derived suspension culture cells in an agitated liquid AA medium. Calli from protoplasts were developed in MS agrose medium containing 2,4- Dlmg/L(The following unit is the same) , KT 0.2 ,NAA 0.2 , Glutamine 867,Asparagine 266. Plantlets were induced on N_6 medium containing KT 2 ,NAA0.2 ,ZT0.2,CH 1000,4%Coconut milk. Regenerated plants were successfully planted in soil.
