摘    要:以建兰品种小桃红根状茎为外植体,研究植物生长调节剂对根状茎增殖、分化和生根的影响,以及根状茎的增殖对培养基中营养物质消耗的变化规律。结果表明,根状茎增殖的最佳培养基是MS+6-BA 0.5mg·L-1+蔗糖30g·L-1,根状茎生长速度为7.33;根状茎分化的最佳培养基是MS+NAA 0.6mg·L-1+TDZ 1.0mg·L-1+蔗糖30g·L-1,分化率为93.10%;生根的最佳培养基是1/2MS+NAA 1.0mg·L-1+蔗糖30g·L-1+AC 0.05%,生根率为96.3%。根状茎增殖的生长量变化呈S型曲线,与细胞培养不同,各时期界限并不是很明显。培养基中碳源、氮源和磷酸盐的消耗曲线与根状茎的生长曲线基本一致。pH由于根状茎先消耗碱性盐(铵态氮)而下降,后利用硝态氮而上升。更多还原

Abstract:The effect of different hormone sort and concentration on multiplication,differentiation and rooting of the rhizome of Cymbidium ensifolium was investigated,The variation of nutrient consumption during the rhizome multiplication was recorded as well.The results indicated that the best condition for rhizome multiplication was the MS+0.5 mg·L-1 6-BA+30 g·L-1 Sucrose,with the growth rate being 7.33;while the MS medium containing 0.6 mg·L-1 NAA,1.0 mg·L-1 TDZ and 30 g·L-1 Sucrose could be optimized for differentiation of rhizome,whose differential rate reached 93.10%.The 1/2 MS supplemented with 1.0 mg·L-1 NAA,30 g·L-1 Sucrose and 0.05 active carbon became the best rooting medium,in which rooting rate reached 96.3%.The growth change of rhizome proliferation was S-shaped curve.Boundaries were not clearly in various periods,which was different from the cell suspension culture system.The consumption of carbon,nitrogen and phosphorus were consistent with those in the rhizome growth curve.The pH of medium firstly decreased and then increased,for the sequence of consumption of ammonium and nitrate.
