摘    要:基于抗原抗体识别特异性,以多巴胺-Mn/ZnS量子点(DA-QDs)为反应媒介,紫外可见分光光度计为检测手段,采用柠檬酸钠还原法制备金纳米颗粒(AuNPs),其表面用辣根过氧化物酶(HRP)和羊抗小鼠免疫球蛋白G (IgG)修饰获得功能化二抗(IgG-AuNPs-HRP)作为信号放大标签,构建一种简便高灵敏的检测方法....

Abstract:Based on the specific interaction between antigen and antibody,a method for detection of alphafetoprotein was developed by using dopamine-Mn/ZnS quantum dots(DA-QDs) as reaction medium and UV-Vis spectrophotometer as detection means.Gold nanoparticles(Au NPs) were prepared by sodium citrate reduction method,and their surfaces were modified with horseradish peroxidase and goat anti-mouse immunoglobulin G to obtain a functionalized secondary antibody(IgG-AuNPs-HRP) as signal amplification label.Under optimized conditions,the response signal had a good linear relationship with the AFP concentration.The linear range was0.1-80 pg/m L,and the detection limit was 0.044 pg/m L.The recovery rates of human serum albumin ranged from 87.7% to 111.5% with the relative standard deviations of 0.2%-1.4%.
