Abstract: | A particularly reliable N-terminal sequence analysis of peptides can be achieved by the following combined application of Edman's phenylisothiocyanate technique, and thin-layer chromatography. The N-terminal amino acids acids split off are identified as phenylthiohydantoin- and dimethylaminonaphthalensulfonyl-derivatives; additionally, quantitative analysis of amino acids using thin-layer chromatography is carried out before and after each degradation step. The applicability of this combined procedure is demonstrated in the case of a nonapeptide. |
