灰毛大青黄酮苷的分析方法和提取工艺

摘    要:采用紫外分光光度法、硝酸铝-亚硝酸钠法和高效液相色谱法分别测定了灰毛大青黄酮苷含量,通过正交试验研究了灰毛大青黄酮苷提取工艺.试验结果表明3种分析方法结果相差较大,通过测定机理分析,确立了硝酸铝-亚硝酸钠法为灰毛大青黄酮苷含量测定的适宜方法,3种方法都具有较好的线性关系,精密度、稳定性、重现性和回收率均符合要求;正交试验的适宜工艺条件为料液比1∶6 (g/V)、提取温度70℃、提取时间60min和提取次数3次,在此条件下黄酮苷得率为9.9330mg/g.

Abstract:The content of flavonoids glycoside in Clerodendrum canescens were determined using ultraviolet ,spectroscopy, aluminum nitrate-sodium nitrite method and high performance liquid chromatography, the extraction process of flavonoids glycoside in Clerodendrum canescens was optimized by means of orthogonaltest design. The experimental results showed that the determination results of content of flavonoids glycoside by the three methods are different from each other. The analysis on mechanism of admeasurement of three methods indicated that the aluminum nitrate- sodium nitrite method is suitable for the determination of flavonoids glycoside in Clerodendrurn canescens, good correlationships, precision, stability, repeatability and recovery were suggested when comparing data obtained by three methods. The orthogonaltest design results showed that the extraction rate of flavonoids glycoside is 9 . 9330mg/g under the optimal conditions of ratio of extractant to raw material 1: 6,extraction temperature 70℃,extraction time 60rain and extraction times 3.
