Abstract:The aim of this study was to explore the microscopic morphology of plant leaves by scanning electron microscopy(SEM), and to find a new method for rapid preparation of SEM leaf samples using tert-butanol. In this study, wild-type Arabidopsis leaves were prepared by preset experimental steps as the research object. Regarding the experimental process and based on the observation results, optimal experimental treatment conditions were summarized as a new SEM rapid sample preparation method for tert-butanol. The results indicated that there were differences in the microscopic morphology of leaf epidermis under different conditions of tert-butanol treatment temperature, duration, freezing time and gold-spraying conductive treatment, among which tert-butanol treatment for 15 min at 30 ℃, freezing on ice for 10 min, and gold-spraying 80 s had the best effect. In this study, a new method for rapid sample preparation of Arabidopsis thaliana leaves by SEM tert-butanol was established, and the experimental results showed that this method also had good observation effect on other species such as Dioscorea oppsita.
