光固定EGF对CHO-K1细胞生长的促进

摘    要:用活化的4-叠氮苯甲酸与上皮细胞生长因子EGF反应,合成了具有光反应性的EGF。将这种EGF涂覆在组织培养聚苯乙烯基板上,通过紫外光照射,EGF与聚苯乙烯基板通过共价键连接。在此材料上进行中国仓鼠卵巢细胞CHO-K1的无血清培养,并对细胞生长活性进行研究。

Abstract:Photo-reactive Epidermal Growth Factor (EGF) was synthesized by coupling EGF with (N-(4-azidobenzoyloxy)succinimid and coated onto the 24-well cell culture polystyrene plate,then Ultraviolet light was irradiated on the coated surface for chemical fixation of EGF via the conversion of the phenylaaldo group to the highly reactive phenyl-nitrene which spontaneously formed covalent bonds with neighboring hydro-carbons. on these EGF-immobilized polystyrene plates, the Chinese harnster ovary (CHO-K1) cell were cultured under the serum-free conditions, and the cell groWth activity were investigated.
