摘    要:通过微弹轰击获得的含有bar基因的转基因燕麦植株自交产生转基因后代。利用PCR方法检测bar基因在后代中的传递情况。

Abstract:Transgenic progenies were obtained by self-pollinating independent transgenic oat (Ayena sativa L.) lines containing har gene driven by rice Actl gene promoter, produced using microprojectile bombardment. Presence of the bar gene was detected in the R1 (the first generation) and R2 (the second generation) progenies of one of the transgenic lines using the Polymerase Chain Reaction, and was confirmed using dot blot hybridization. A Mendelian segregation was found in all the R2 populations examined, but not in the R1 progeny, which was probably caused by sampling error. One of the R2 populations was found to be uniformly bar gene positive, indicating it being derived from a homozygous R1 plant. A stable expression of the bar gene was detected in both the R1 and R2 progenies. No inactivation of bar gene expression was observed in the progeny populations examined.
