With an F2 population from the cross of two cucumber inbred lines, S06 and S52, sequence-related amplified polymorphism (SRAP) was used to construct a genetic linkage map in cucumber (Cucumis sativus L.). Sixty-four SRAP primer combinations generated 108 polymorphic bands in the F2 population analysis. The average of polymorphic bands produced by one primer pair was 1.5, and the maximum was 5. Using Mapmaker 3.0, a linkage map was constructed, which consisted of 77 SRAP markers distributed in nine linkage groups (LOD≥3.0) and spanned 1114.2 cM with an average interval of 14.5 cM between markers. The gene for the first-flower-node trait, termed ffn, was mapped to linkage group Ⅸ, flanked by DC1EM5 and ME7EM2A at 10.3 cM and 12.1 cM distance, respectively. 相似文献

以4-氯酚(4-CP)、2,4-二氯酚(2,4-DCP)和2,4,6-三氯酚(2,4,6-TCP)为模型化合物,建立了中空纤维膜支载-可忽略损耗液相微萃取法(HF-nd-LPME)和高效液相色谱(HPLC)联用,同时测定辛醇-水分配系数(Kow)和电离常数(pKa)的方法。在60 mL正辛醇饱和的样品溶液(1mmol/L NaH2PO4,2μg/mL4-CP、2,4-DCP和2,4,6-TCP)中放入2个支载有0.16μL正辛醇的中空纤维膜萃取装置,静置96 h确保达到萃取平衡。然后将萃取装置取出并用甲醇解吸,经HPLC测定得到萃取到正辛醇中的目标物浓度coctanol。coctanol与样品中目标物原始浓度cwater的比值即为Kow。通过测定模型化合物在不同pH3.0~13条件下的Kow值,并依据化合物的Kow、pKa与pH的关系模型进行非线性拟合,即可计算得出模型化合物pKa值。实验表明,本方法测定的三种模型化合物的Kow和pKa值与文献报道值一致。 相似文献

This article presents a new, simple and rapid continuous separation method by combination of flow injection with capillary electrophoresis designed for the analysis of basic traditional Chinese medicines. The device was produced using commercial capillary and components readily available in analytical laboratory. In double-T configuration, the designed horizontal separation channel was 25 microm i.d. x 146 mm length (an effective separation length of 93 mm) quartz capillary, with two vertical elicitation arms produced from 0.5 mm i.d. pump tubing. The capillary was embedded in a 40 x 20 x 3 mm organic glass base. Using the double-T configuration, continuous introduction of a series of samples was achieved. More than 3.00 resolution for ephedrine and pseudo-ephedrine were obtained using 100 mm borate buffer (pH 9.80) within 8 min in 25 microm separation channel with an electrical field strength of 137 V/cm (UV detection at 215 nm). The linear calibration range was 50-1500 microg/mL (ephedrine, r = 0.9982; pseudo-ephedrine, r = 0.9990) for both analytes. The limits of detection were 2.65 micro g/mL for ephedrine and 2.92 microg/mL for pseudo-ephedrine. In this device, the contents of ephedrine and pseudo-ephedrine in five Chinese medicinal preparations were determined with RSDs (n = 5) in range 1.16-4.51% and recoveries in range 90.4-114.6%. 相似文献

In this paper the structure correlation of PE-PS gradient polymers, modified by styrene LDPE films with their mechanical and rheo-optical properties, is presented. The specific structural changes of both the investigated materials can be demonstrated by using dynamic mechanical deformation studies. The influence of the penetrant on LDPE crystalline and amorphous phase order is shown. Some interesting mechanical behaviour of the gradient polymers, depending on the polystyrene contents, is discussed. 相似文献
