A total of 27 yeast strains belonging to the groupsCandida, Saccharomyces, andKluyveromyces were screened for their ability to grow and ferment glucose at temperatures ranging 32-45°C. K. marxianus andK. fragilis were found to be the best ethanol producing organisms at the higher temperature tested and, so, were selected for subsequent simultaneous saccharification and fermentation (SSF) studies.  相似文献

One of the limiting factors restricting the effective and efficient bioconversion of softwood-derived lignocellulosic residues is the recalcitrance of the substrate following pretreatment. Consequently, the ensuing enzymatic process requires relatively high enzyme loadings to produce monomeric carbohydrates that are readily fermentable by ethanologenic microorganisms. In an attempt to circumvent the need for larger enzyme loadings, a simultaneous physical and enzymatic hydrolysis treatment was evaluated. A ball-mill reactor was used as the digestion vessel, and the extent and rate of hydrolysis were monitored. Concurrently, enzyme adsorption profiles and the rate of conversion during the course of hydrolysis were monitored. α-Cellulose, employed as a model substrate, and SO2-impregnated steam-exploded Douglas-fir wood chips were assessed as the cellulosic substrates. The softwood-derived substrate was further posttreated with water and hot alkaline hydrogen peroxide to remove >90% of the original lignin. Experiments at different reaction conditions were evaluated, including substrate concentration, enzyme loading, reaction volumes, and number of ball beads employed during mechanical milling. It was apparent that the best conditions for the enzymatic hydrolysis of α-cellulose were attained using a higher number of beads, while the presence of air-liquid interface did not seem to affect the rate of saccharification. Similarly, when employing the lignocellulosic substrate, up to 100% hydrolysis could be achieved with a minimum enzyme loading (10 filter paper units/g of cellulose), at lower substrate concentrations and with a greater number of reaction beads during milling. It was apparent that the combined strategy of simultaneous ball milling and enzymatic hydrolysis could improve the rate of saccharification and/or reduce the enzyme loading required to attain total hydrolysis of the carbohydrate moieties.  相似文献

With the object to synthesize polyesters by enzymatic catalysis in organic media, two directions have been investigated: (1) the condensation polymerization of linear ω-hydroxyesters and (2) the ring-opening polymerization of lactones. The commercially-available crude porcine pancreatic lipase (PPL), suspended in organic solvents, was the preferred enzyme for the reactions. In order to determine the optimal conditions for the condensation polymerization, the bifunctional methyl 6-hydroxyhexanoate was used as a model compound to study the influence of the following parameters: type of the enzymecatalyst, kind of solvent, concentration, temperature, duration, size of the reaction mixture, and stirring. Film-forming polyesters with a degree of polymerization (DP) up to about 100 were obtained from linear aliphatic hydroxyesters in n-hexane at reflux temperature (69°C). Yet concurrently with the intermolecular condensation polymerization, macrolactones were also formed by intramolecular reaction. Two aromatic hydroxyesters did not react under these conditions. For the ring-opening polymerization of lactones the reaction of ?-caprolactone with methanol as the preferred nucleophile, was studied. Polyesters with a DP of up to 35 were obtained in n-hexane at temperatures between 25 and 40°C. The degrees of polymerization of the polyesters were determined by comparative analyses of the end groups in the 1H-NMR spectra and by determination of molecular weights either by vapor phase osmometry, gel permeation chromatography, or intrinsic viscosity. © 1993 John Wiley & Sons, Inc.  相似文献

Enzyme-catalyzed preparation of polymers offers several potentially valuable advantages over the usual polymerization procedures and has been studied for several years. A significant limitation on the polyesters prepared to date has been the low molecular weights achieved. The present studies have established that, in the polycondensation of bis(2,2,2-trifluoroethyl) glutarate with 1,4-butanediol using porcine pancreatic lipase as the catalyst, this limitation arises from at least two sources: hydrolysis of activated ester end groups by water introduced along with the enzyme and the polymerization's reaching equilibrium despite using the poorly nucleophilic 2,2,2-trifluoroethanol as the leaving group. Evidence is also developed that the presence of trifluoroethanol accelerates the release of the enzyme-bound water which hydrolyzes the activated ester end groups. The hydrolysis could be avoided by choosing a relatively high-boiling solvent, such as bis(2-ethoxyethyl) ether, then removing the trifluoroethanol by placing the reaction mixture under vacuum periodically or by drying the enzyme rigorously. The vacuum method also removed the limitation on molecular weight resulting from the reaction's reaching equilibrium. A further improvement in the molecular weight to nearly 40,000 daltons, well within the range that is technically interesting, was achieved by using 1,2-dimethoxybenzene or 1,3-dimethoxybenzene as the polymerization solvent. © 1995 John Wiley & Sons, Inc.  相似文献

The hydrogen peroxide-oxidation of o-phenylenediamine (OPD) catalyzed by horseradish peroxidase (HRP) at 37 °C in 50 mM phosphate buffer (pH 7.0) was studied by calorimetry. The apparent molar reaction enthalpy with respect to OPD and hydrogen peroxide were −447 ± 8 kJ mol−1 and −298 ± 9 kJ mol−1, respectively. Oxidation of OPD by H2O2 catalyzed by HRP (1.25 nM) at pH 7.0 and 37 °C follows a ping-pong mechanism. The maximum rate Vmax (0.91 ± 0.05 μM s−1), Michaelis constant for OPD Km,S (51 ± 3 μM), Michaelis constant for hydrogen peroxide Km,H2O2 (136 ± 8 μM), the catalytic constant kcat (364 ± 18 s−1) and the second-order rate constants k+1 = (2.7 ± 0.3) × 106 M−1 s−1 and k+5 = (7.1 ± 0.8) × 106 M−1 s−1 were obtained by the initial rate method.  相似文献

Oxidation of lignin obtained from acetosolv and ethanol/water pulping of sugarcane bagasse was performed by phenol oxidases: tyrosinase (TYR) and laccase (LAC), to increase the number of carbonyl and hydroxyl groups in lignin, and to improve its chelating capacity. The chelating properties of the original and oxidized lignins were compared by monitoring the amount of Cu2+ bound to lignin by gel permeation chromatography. The Acetosolv lignin oxidized with TYR was 16.8% and with LAC 21% higher than that of the original lignin. For ethanol/water lignin oxidized with TYR was 17.2% and with LAC 18% higher than that of the original lignin.  相似文献
