Summary The enzymes a-amylase, invertase and glucoamylase were immobilized on acid activated montmorillonite using two techniques,
viz. adsorption and covalent binding, and their activities were tested in a batch and packed-bed reactor and were compared. The
packed-bed reactor showed an improved performance for all immobilized enzymes, which was attributed to lowering of diffusional
restrictions to mass transfer. Lower activity in case of batch reactor for immobilized invertase was due to a combined effect
of loss of native conformation of enzyme on account of immobilization and mass transfer resistances due to improper diffusion
of substrate to the active site of enzyme. For immobilized glucoamylase, the packed-bed reactor demonstrated exceptionally
high activity that was very close to the free enzyme. Covalently bound glucoamylase showed higher activity than the free enzyme. 相似文献

报道了一种新的L-氨基酸氧化酶电极,这种酶电极系由氨气敏电极和以氨基化玻璃布为载体的酶膜所组成;研究了固定化条件对酶膜活性的影响以及底物浓度、温度和pH对电极响应特性的影响。该电极在6.0×10^-5~4.0×10^-3mol/L的底物浓度范围内呈良好的线性关系,检测下限为5.0×10^-5mol/L。在最宜条件下,酶电极具有良好的稳定性。 相似文献

The structure and properties of the blend of regenerated silk fibroin (RSF) and poly(vinyl alcohol) (PVA) were investigated.
The two polymers in the blend are in the state of phase segregation. Infrared (IR) spectra indicate that the RSF in the blend
maintains its intrinsic properties, thus, ethanol treatment can transfer silk I structure of RSF to silk II structure. The
water absorption property and mechanical property of the blend are improved in comparison with those of RSF. The blend maintains
the major merit of RSF, that is, it can immobilize glucose oxidase on the basis of the conformational transition from silk
I structure to silk II structure. The properties of the immobilized enzyme are examined. Moreover, the second generation of
glucose sensor based on the immobilized enzyme is fabricated and it has a variety of advantages including easy maintenance

Increased utilization of mechanically stable synthetic matrices particularly silica gel as a solid support and its surface modification either by impregnation of organic ligands directly or covalent grafting through spacer unit for extractive concentration of trace elements are highlighted in the present article. Experimental evidences for existence of surface silanol and its chemical nature have explored the idea of silica surface modification. Recent methods of development in functionalized silica synthesis by attachment of various ligands or organic reagents to the silica surface and techniques of characterization of the modified surface have been reported. Analytical applications of various modified silica surfaces, in particular, adsorption of trace elements taking separation and preconcentration into account from complex synthetic mixture as well as natural water is presented. 相似文献

本文继采用含硫配体(-SH,-SR)对高分子保护金属胶体的配位俘获之后,采用锚联在SiO_2表面的较含硫配体配位能力弱的P,N配体分别与聚乙烯吡咯烷酮(PVP)保护的胶体钯在室温下反应,在P/Pd大于3.5,N/Pd大于7时,可以实施对保护金属胶体的配位俘获,证实了配位俘获法是一种具有普遍意义的方法,经原子吸收光谱分析,溶液中残留的金属钯的量小于0.36 ppm(P/Pd=3.5)。经电子能谱证实,锚联膦胺配体与金属钯之间存在配位作用,这是实施保护胶体负载化的关键。透射电镜分析表明,在整个配位俘获的过程中,无论是在载体上或是在溶液中,胶体钯的颗粒大小与分布均保持不变。因此,配位俘获法是一种能控制多相催化剂中金属颗粒大小与分布的有效方法。由此合成的钯催化剂具有很高的催化活性,良好的选择性和稳定性,在对Cis,Cis-1,3-环辛二烯的选择性加氢中,其活性仅比均相的PVP-Pd胶体催化剂稍低,选择性达100%。在周转数达12,000次/个钯原子后,其催化活性仍保持不变,随着P/Pd比的增大,催化活性迅速下降。 相似文献

A new detection system based on microdialysis sampling and chemiluminescence (CL) reaction was developed for in vivo monitoring of uric acid (UA) with high sensitivity, selectivity and accuracy. The uric acid is indirectly monitored by CL detection of enzymatic reaction product formation (H2O2), catalyzed by Uricase. A microprobe was modified and coated with immobilized enzyme through a Streptavidin-biotin mediated linker by using a chitosan support membrane, polyurethane trapped ferrocene film is employed to protect the probe surface and diminish the interference from reductant molecules, which often are present in the blood (e.g. ascorbic acid). The earlier mentioned probe and the constructed sensor can detect uric acid in the range of 0.01-1 mM with detection limit (3σ) of 5 μM. Finally, the system is used to monitor uric acid (UA) variation through an acute myocardial infarction (AMI) model. Following AMI-induced oxidative stress, the UA level decreases continuously, thus suggesting that UA plays a protective role as a substitute antioxidant. Furthermore, the in vivo monitoring results show good agreement with those obtained by a standard method, and the procedure is recommended for in vivo and real time monitoring of UA. In addition, the proposed method can be more accurate since the UA may be potentially oxidized by in vitro exposure to oxygen in the presence of a catalyst. 相似文献

The immobilization of copper in organic-inorganic hybrid materials catalyzing the Ullmann reaction has been described. Phenols reacted with aryl iodides, aryl bromides and aryl chlorides smoothly in the presence of a 3-(2-aminoethylamino)propyl functionalized silica gel immobilized copper catalyst. The protocol involved the use of DMSO as the solvent, and potassium fluoride as the base. The reactions generated the corresponding cross-coupling products in good to excellent yields. Furthermore, the silica-supported copper could be recovered and recycled by a simple filtration of the reaction solution and used for 10 consecutive trials without loss of its reactivity. 相似文献
