目的:建立用高效液相色谱(HPLC)法测定小儿导赤片中大黄素的含量.方法:采用HPLC法,色谱柱为Agilent ZORBAXSB-C18(4.6mm×150 mm,5μm),流动相为甲醇-0.1%磷酸水溶液(80:20),流速为1.0 ml/min,柱温为30℃,检测波长为254nm.结果:大黄素在0.04~0.4μg(r=0.999 9)范围内有良好的线性关系.大黄素平均回收率分别为98.25%.结论:该方法操作简便、快速、结果准确可靠. 相似文献

A sensitive and rapid Spectrofluorimetric method for the determination of guanethidine sulphate, guanfacine hydrochloride, guanoclor sulphate and guanoxan sulphate in tablets and spiked human serum and urine samples is described. The method is based on the reaction of monosubstituted guanidino compounds in an aqueous potassium hydroxide solution with benzoin, in the presence of -mercaptoethanol and sodium sulphite. Highly fluorescent derivatives were obtained, with excitation and emission maximum wavelengths around 325 and 430 nm, respectively. In optimal reaction conditions, the linearity ranges were 0.04–0.28 g/ml, with relative standard deviations less than 2%. The method has been successfully applied to the determination of these drugs in tablets. The results are highly correlated with the B.P. method. Chloroform (or for guanoxan dichloromethane) was used to extract the drugs from serum and urine at basic pH, followed by the proposed fluorimetric method. The limit of detection is 0.02 g/ml for the selected drugs. 相似文献

根据吸光度比-光谱曲线,选定215.7 nm及264.1 nm作为测定氯唑沙宗(CRXZ)和对乙酰氨基酚(ACAP)的波长对。制备标准曲线时,首先配制由CRZX和ACAP基准物配制的标准溶液和由两者配成的混合标准系列,并在上述两波长处分别测得各试液的吸光度,再用文中提供的公式计算吸光度比值R和吸光度之差ΔR,最后建立ΔR对CRZX或ACAP浓度之间的线性回归方程。分析试样时,取复方氯唑沙宗片适量溶于溶剂中并定量稀释。分取部分试液按制备标准曲线的方法测得试液的吸光度并计算其比值R和ΔR。根据相应回归方程计算相应组分的含量。精密度及回收率试验所得结果的RSD(n=7)值为1.28%(CRXZ)及0.84%(ACAP),两者的平均回收率均为100.0%。 相似文献

This paper describes the validation of an isocratic HPLC method for the assay of voriconazole in tablets. The method employs
a Merck LiChrospher? 100 RP-8 (125 × 4.6 mm I.D., 5 μm particle size) column, with a mobile phase of methanol : triethylamine solutions 0.6 %,
pH 6.0 (50:50, v/v) and UV detection at 255 nm. A linear response (r > 0.9999) was observed in the range of 20.0–100.0 μg mL−1. The method showed good recoveries (average 100.4%) and the relative standard deviation intra and inter-day were ≤ 1.0 %.
Validation parameters as specificity and robustness were also determined. The method can be used for both quality control
assay of voriconazole in tablets and for stability studies as the method separates voriconazole from its degradation products

In this study, a fingerprint-activity relationship modeling between chemical fingerprints and antirheumatic activity was established, and multivariate statistical analysis was used to evaluate the quality of Taxilli Herba (TH) from different hosts. Characteristic fingerprints of 20 batches of TH samples were generated by high-performance liquid chromatography coupled with triple quadrupole-time of flight tandem mass spectrometry (HPLC-Triple TOF-MS/MS), and the similarity analysis was calculated based on thirteen common characteristic peaks by hierarchical clustering analysis (HCA). Subsequently, nine efficacy markers were discovered by combining fingerprints and antirheumatic activity through grey correlation analysis (GCA) and bivariate correlation analysis (BCA). Meanwhile, the content of 5 constituents in 9 markers was determined by high-performance liquid chromatography coupled with triple quadrupole-linear ion trap tandem mass spectrometry (HPLC-QTRAP-MS/MS). The comprehensive quality of TH was assessed using multivariate statistical analysis, including principal components analysis (PCA) and technique for order preference by similarity to ideal solution (TOPSIS). The results showed that a high dose of TH extract could markedly ameliorate arthritis damage compared to other doses, with flavonoids playing an important role in the antirheumatic activity. The comprehensive quality of samples from Morus alba L. (SS) was superior to those from Liquidambar formosana Hance (FXS). The present study will demonstrate the markers associated with efficacy, and provide an applicable strategy for more comprehensive quality control and evaluation of TH. 相似文献

By means of spectroscopic methods it has been found that demeclocycline reacts with molybdate ions forming a complex compound [MoO3HDMTC]2–. The relative stability constant of this compound has been determined by applying spectrophotometric methods. Under optimum conditions for complex formation a very sensitive spectrophotometric method for the estimation of demeclocycline in the concentration range 5.0–35.1 g/ml is proposed. The detection limit of the method is 2.5 g/ml of demeclocycline. The relative standard deviation (n=10) varies between 0.76% and 1.46%. The method proved to be accurate and sensitive for the analysis of the demeclocycline content in tablets. 相似文献

Colorimetric and Spectrofluorimetric Methods for the Determination of Melatonin in Tablets and Serum
Abstract Two sensitive and accurate colorimetric and spectrofluonmetric methods, are presented for the determination of melatonin in tablets and serum. The first method utilizes the reactions of p-dimethylaminobenzaldehyde in hydrochloric acid (van Urk reagent)-ferric chloride in sulphuric acid (Salkowski reagent) mixture. The blue color of the resulting reaction product is measured at 630 nm. The second method is based on the reaction of melatonin with o-phthalaldehyde in acid medium which yields highly fluorescent condensation product that is measured at 465 nm as emission wavelength, using excitation wavelength at 355 nm. No interference was observed from tableting additives, and the applicability of the methods was examined by analysing tablets containing melatonin (single and combined with pyridoxine). Mean percentage recoveries from tablets were found to be 99.9+0.31 for single and 100.5+0.15 for combined tablets using colorimetric method, while by applying spectrofluorimetric method the recoveries were found to be 100.610.41 for single and 100.2+0.39 for combined tablets. Furthermore, the proposed methods were extended to the in-vitro determination of melatonin in serum. The detection limits are 0.27 ug ml?1 for colorimetric method and 0.00035 ug ml?1 for spectrofluorimetric method. 相似文献
