本文研究了枯草杆菌AS1.398发酵培养制备中性蛋白酶,对发酵培养基配比,培养条件,金属离子的影响等进行了摇瓶试验。制备的AS1.398中性蛋白酶有如下性质:最适PH7.2-7.4,在PH6.5-8.0稳定。最适温度42-44℃,35℃下处理2小时能保持约85%酶活力,60℃下10分钟酶完全失活,此酶被EDTA和Cu2+所抑制  相似文献

通过实验室饲料感染方法和大田喷杀对比试验研究,表明发酵培养基中添加花生饼可以提高HD—1的杀虫效果,且添加浓度与HD—1菌株毒力呈正相关,用添加了花生饼的固体浅盘发酵培养液比市售工业化生产的同类杀虫剂杀虫效果要高得多。  相似文献

The first objective of this study was the measurement of physical properties of P(3HB-co-3HV) copolymers with different (hydroxybutyrate) HB to (hydroxyvalerate) HV ratios produced by Bacillus cereus (TRY2) isolated from activated sludge. The 3HV PHBV copolymers were 0.05, 22.6, 39.2, 54.1, and 69.1 mol%, respectively. The second objective was to study possible wastewater treatment and production of PHAs at the same time by B. cereus (TRY2) and Pseudomonas spp. (TOB17) (both were isolated from activated sludge), recombinant Bacillus DH5α, and a combination of the above three bacteria. The results were satisfactory; the maximum COD and TOC of the sewage sludge reduced were 53.5% and 67.5%, respectively.  相似文献

The application of Bacillus subtilis as a flocculant for fine coal has been reported here. Zeta-potential measurements showed that both the coal and bacteria had similar surface charge as a function of pH. Surface free energy calculations showed that the coal was hydrophobic while the bacterium was hydrophilic. The adhesion of the bacteria to coal and subsequent settling was studied in detail. Adhesion of bacteria to coal surface and subsequent settling of coal was found to be quick. Both adhesion and settling were found to be independent of pH, which makes the process very attractive for field applications. The presence of an electrolyte along with the bacterium was found to not only enhance adhesion of bacteria, but also produce a clear supernatant. Further, the settled fraction was more compact than with bacteria alone. Interaction energy calculations using the extended DLVO theory showed that the electrical forces along with the acid–base interaction energy play a dominant role in the lower pH range. Above pH 7, the acid–base interaction energy is the predominant attractive force and is sufficient enough to overcome the repulsive forces due to electrical charges to bring about adhesion and thus settling of fine coal. With increase in electrolyte concentration, the change in total interaction energy with pH is minimal which probably leads to better adhesion and hence settling.  相似文献

This article reports studies concerning the production of penicillin G acylase (PGA) by Bacillus megaterium. This enzyme has industrial use in the hydrolysis of penicillin G to obtain 6-aminopenicillanic acid, an essential intermediate for the production of semisynthetic β-lactam antibiotics. Although most microorganisms produce the enzyme intracellularly, B. megaterium provides extracellular PGA. The enzyme production by microorganisms involves several steps, resulting in a many operational variables to be studied. The study of the inoculum is an important step to be accomplished, before addressing other issues such as culture optimization and downstream processing. In this study, using a standard inoculum as reference, several runs were performed aiming at the definition of operational conditions in the PGA production. Cell concentration and PGA activity in the production medium were measured after 24, 48, and 72 h of the beginning of the production phase. This study encompasses the duration of the inoculum germination phase and the concentration of cells used to startup the germination. Based on these results, PGA productivity during the production phase was maximized. The selected values for these variables were 1.5 × 107 spores/mL of germination medium, germination during 24 h, and 72 h for the production phase.  相似文献

Thermostable α-amylases have application in a variety of industrial processes and enzymes from a substantial number of thermophilic bacteria and fungi have been screened and characterized to varying degrees. The characteristics of these enzymes are summarized in this review. The genetics of α-amylase production inBacillus subtilis is reviewed and classical and recombinant DNA approaches to increasing α-amylase production are discussed.  相似文献

Spectra obtained using matrix-assisted laser desorption/ionization (MALDI) mass spectrometry of Bacillus globigii (Bacillus subtilis niger) spores, vegetative cells and the culture supernatant show a cluster of biomarkers centered at a molecular mass of 1478 Da. Three biomarkers were isolated from the cell-free culture supernatant by solid-phase extraction and reversed-phase high-performance liquid chromatography, and characterized using various kinds of mass spectrometry. A Fourier transform mass spectrometer with a MALDI source was used to determine the monoisotopic protonated masses at 1463.8, 1477.8, and 1505.8 Da in order of elution. The mass differences of 14 and 28 Da suggest that they are homologous molecules. Alkaline hydrolysis of each species showed that it contained a lactone linkage. Strong acid hydrolysis released a fatty acid from an amide bond, consistent with a lipopeptide. A quadrupole time-of-flight instrument with a nanospray source was used to sequence the hydrolyzed forms of the three biomarkers. The cyclic lipopeptides were found to have amino acid sequences identical with those in fengycins and plipastatins, antimicrobial compounds with phospholipase inhibitor activity, previously identified in related species of Bacillus subtilis and Bacillus cereus.  相似文献

Large-volume parenteral solutions were submitted to heat treatments after being inoculated with Bacillus stearothermophilus ATCC 7953 (T r =121°C) and Bacillus subtilis ATCC 9372 (T r =104.5°C) spores. The average decimal reduction time for B. stearothermophilus ranged from a D 121°C value of 1.31 to 3.14 min, in glucophysiologic and Ringer’s solutions respectively. For B. subtilis, D 104.5°C value increased from 0.69 to 1.37 min, in Ringer’s (pH=5.91) and 50% glucose (pH 3.05) solutions respectively. The z value ranged from 7.95°C (20% mannitol solution) to 13.14°C (50% glucose solution), corresponding to an activation energy (Ea) of 81.48 and 49.30 kcal/mol, respectively.  相似文献

试验证实,嫩毛竹自然发酵过程中的微生物主要是细菌。分离出并纯化了100株细菌,其中60株是芽胞杆菌。将分离出的100株细菌分别接入石灰预处理过的嫩竹,从中筛选出能使嫩竹进一步软化的菌株3株,也均为芽胞杆菌。对这60株芽胞杆菌进行了形态学及其生理生化特征的鉴定,按Priest氏等(1988年)的数值分类系统分类到组,将3株(308,309,310)对嫩竹软化有明显作用的芽胞杆菌的1株(308)鉴定到种,认为是短小芽胞杆菌(B.pumilus)。另外40株非芽胞杆菌参阅《伯杰氏系统细菌学手册》第1卷和第2卷,鉴定到科或属,有微球菌、葡萄球菌、李氏杆菌、不动杆菌、肠杆菌科等。  相似文献

The growth ofBacillus amyloliquefaciens in the aqueous two-phase system, made up of polyethylene glycol, dextran, and water, was investigated. Generally,Bacillus partitions in the dextran phase, but the magnitude of the separation depends largely on the overall composition of polymers in the phase system. The kinetics of growth ofBacillus amyloliquefaciens was studied in the polyethylene glycol-rich continuous phase, dextran-rich dispersed phase, and in the mixed phase. From the kinetic data it appears that increasing the overall polymer composition causes the cells to adsorp at the interface. On the other hand, partition measurements indicate that increasing polymer concentrations make the cell partitioning more one-sided. This anomaly is explained by studying the interfacial adsorption of cells via dynamic surface tension measurements.  相似文献
