Summary The acidity constants of adenosine-5-mono- and diphosphate (AMP andADP) were determined at 25.00±0.1°C by potentiometric titration in pure water and different solvent mixtures (methanol, ethanol, N,N-dimethylformamide, dimethylsulfoxide, acetone, and dioxane). The ionization ofAMP andADP depends on both the proportion and the nature of the organic solvent used. ThepK a1 values for bothAMP andADP are slightly influenced as the solvent is enriched in ethanol and methanol and remains practically constant in presence of different amounts ofDMF andDMSO. A pronounced change in thepK a1 values is observed as the solvent is enriched in acetone or dioxane. It is concluded that the electrostatic effect has only a relatively small influence on the dissociation equilibrium, whereas other solvent effects such as solvent basicity, hydrogen bonding and protonsolvent interactions play an important role.

Zusammenfassung Die Aciditätskonstanten von Adenosin-5-mono- und -diphosphat wurden bei 25.0±0.1°C in reinem Wasser und in verschiedenen Lösungsmittelgemischen (Methanol, Ethanol, N,N-Dimethylformamid, Dimethylsulfoxid, Aceton und Dioxan) potentiometrisch bestimmt. Der Ionisierungsgrad vonAMP undADP hängt sowohl von der Menge als auch von der Art des organischen Lösungsmittels ab. DiepK a1-Werte vonAMP undADP werden durch Zugabe von Methanol und Ethanol nur wenig, durch verschiedene Mengen vonDMF undDMSO gar nicht, durch Aceton und Dioxan jedoch deutlich beeinflußt. Offensichtlich haben elektrostatische Effekte nur geringe Auswirkungen auf das Dissoziationsgleichgewicht, wogegen andere Faktoren wie Basizität des Lösungsmittels, Wasserstoffbrückenbindungen und Lösungsmittel-Proton-Wechselwirkungen eine bedeutende Rolle spielen.

三七叶甙对兔血小板聚集功能的影响   总被引:4,自引:1,他引:4  
 应用Born氏比浊法分别测定三七叶甙(Panax notoginseng folium saponins)对花生四烯酸(arachidonic acid,AA)、血小板活化因子(platelet activating factor,PAF)和腺苷二磷酸(adenosine diphosphate,ADP)诱导的兔血小板聚集功能的影响.实验结果表明:三七叶甙在体外显著抑制ADP诱导的血小板聚集,其半数抑制浓度(medium inhibitory concentration,IC50)为89.4mg/L;三七叶甙静脉注射也能明显降低ADP诱导的兔血小板聚集率,且呈剂量-效应关系.提示三七叶甙体内外均明显抑制ADP诱导的兔血小板聚集,而对AA和PAF诱导的兔血小板聚集无明显影响.  相似文献

Pure and thiourea substituted single crystals of ammonium di-hydrogen phosphate have been grown from aqueous solution by isothermal solvent evaporation technique. Doped crystal exhibits prominent changes in physical and chemical properties. Single crystal XRD analyses of the samples are carried out and the results are compared. FTIR and UV–vis–NIR spectral analyses have been employed to identify the presence of various functional groups and the UV cut-off range in the grown crystals. Density measurements have been made and Photoconductivity studies revealed the negative photo conducting nature. Hardness measurement shows that the mechanical strength of the doped crystal is high when compared to pure ammonium di-hydrogen phosphate. The dielectric response of the samples has been studied in the frequency range 100 Hz–5 MHz at room temperature and the results are discussed.  相似文献

A UHPLC–MS/MS method for the quantification of ADP355, an adiponectin-derived active peptide, was developed and validated. The extraction method employed simple protein precipitation using methanol and chromatographic separation was achieved on anAccucore™ RP-MS C18 column (100 × 2.1 mm, 2.6 μm, 80 Å), using 0.1% formic acid in both water and acetonitrile with gradient elution at the flow rate of 400 μl/min within 4.0 min. Detections were performed under positive ion mode with multiple reaction monitoring ion transitions m/z 1109.2 → 309.8 and 871.4 → 310.1 for ADP355 and Jt003 respectively at unit resolution. The linearity range of the calibration curve was 2–1,000 ng/ml with a lower limit detection of 0.5 ng/ml. The selectivity, linearity, precision, accuracy, recovery, matrix effect and stability were validated, and all items met the requirement of US Food and Drug Administration guidance. This method was successfully applied to an intravenous pharmacokinetic study of ADP355 in rats and the in-vitro stability in rat serum, plasma and whole blood was also assessed.  相似文献

Ammonium dihydrogen phosphate NH4H2PO4 (ADP) is an antiferroelectric (AFE) compound belonging to the KDP-type family of hydrogen-bonded ferroelectrics. Recent ab initio results have shown that the optimization of the N–H–O bridges in ADP leads to the stabilization of the AFE state over a FE one. However, electron spin probe measurements have suggested that microregions of both phases may coexist above the critical antiferroelectric–paraelectric transition temperature. We have performed first principles studies of the energetics and relative stability of different AFE and FE defects embedded in a paraelectric (PE) matrix of ADP. Our analysis indicates that FE and AFE clusters are stable and may coexist in the PE phase, thus confirming the above suggestion.  相似文献

ADP (NH4H2PO4) crystals were grown through the traditional method and rapid growth technology. The optical homogeneity of rapid grown ADP crystals was analyzed by laser interferometry and X-ray topography. Laser interferometry revealed sector boundaries at which optical homogeneity decreased dramatically. The main defects that seriously reduced the optical homogeneity of the rapid grown ADP crystal were sector boundaries, growth bands and inclusions. The concentration of Fe and Cr impurities was tested by a plasma emission spectrometer. It was found that the preferential incorporation of metallic impurities into the prismatic faces resulted in high density of growth bands and inclusions, thus reducing optical homogeneity of the prismatic sector.  相似文献
