A combined-modality treatment program consisting of botulinum toxin injection (Botox) and voice therapy was used to treat 17 subjects diagnosed with adductor spasmodic dysphonia (ADD SD). Ten subjects with ADD SD served as the control and were given Botox only. Voice therapy after Botox injection was directed toward reducing the hyperfunctional vocal behaviors, primarily glottal overpressure at voice onset and anterior-posterior squeezing. The results indicated that subjects who underwent combined-modality treatment maintained significantly higher mean airflow rates for significantly longer periods. Moreover, there was a carryover effect in these patients when they received Botox only. Adductor spasmodic dysphonia is treated most effectively when intrinsic laryngeal muscle spasms are reduced or eliminated by Botox injection and extrinsic hyperfunctional vocal behaviors are treated with voice therapy  相似文献

建立了免疫亲和柱净化/柱前衍生化-高效液相色谱荧光检测器测定粮谷中T-2毒素含量的方法。样品经甲醇-水(体积比为80∶20)混合溶剂提取,通过免疫亲和柱(IAC)净化,以氰酸蒽(1-AN)为衍生化试剂、4-二甲基氨基吡啶(DMAP)为催化剂进行衍生,以ZORBAX Eclipse XDB-C18 柱为分离柱,乙腈-水(体积比为80∶20)为流动相进行高效液相色谱分离及荧光检测,荧光检测的激发波长为381 nm,发射波长为470 nm。T-2毒素的质量浓度为0.01~1.5 mg/L时与峰高呈良好的线性,相关系数为0.9985。在0.01~1.5 μg/g添加水平下,回收率为79.7%~94.5%,相对标准偏差小于7%;检出限(S/N=3)为0.01 μg/g。该方法净化效果好,灵敏度高,操作简便快速。  相似文献

Surface plasmon resonance (SPR) has been successfully applied for the simple, rapid, and label-free assay of various biomolecules. This assay evaluates a novel wavelength modulation SPR biosensor for the detection of tetanus toxin. The wavelength modulation SPR biosensor is designed based on fixing the incident angle of light and measuring the reflected intensities in the resonance wavelength range spanning 400-800 nm simultaneously. Tetanus toxin (TeNT), one of the most potent toxins known, is synthesized as a 150 kDa single polypeptide chain. The SPR biosensor has been shown to be capable of directly detecting concentration of tetanus toxin as low as 0.028 Lf ml−1. Under selected experimental conditions, the SPR biosensor has a good reproducibility, sensitivity and reversibility. The results illustrate how wavelength modulation SPR biosensor can be used to detect biomolecular interactions.  相似文献

Microorganisms produce toxins against its competitors sometimes, and variable yields are useful to explain the observed oscillatory behavior in the reactor. In this paper, a model with general quadric yields of competition in the bioreactor of two competitors for a single nutrient where one of the competitors can produce toxin against its opponent, is proposed. We analyze the asymptotic behavior of the model in terms of the relevant parameters. The conditions of the three dimensional Hopf bifurcation, and the existence of limit cycles in the nutrient-organism phase plane are obtained.  相似文献

A solid-phase route for synthesis of 15N-labeled acylpolyamines is described. Utilizing alkylation at benzylic N-atom as a key step, 15N-atoms are incorporated by stepwise construction of the polyamine framework on the solid support. We report the fabrication of a microfluidic apparatus and the realization of a sensors based on PEDOT : PSS, a biocompatible semiconductor polymer used in substitution of standard electrodes for electrophysiological studies and for detection of nanopores in membrane. This gives the possibility to study the mechanisms of ions balance and molecular transport though cell membranes. In particular the apparatus is based on two chambers connected through an aperture in a PTFE sheet where lipid bilayer are formed using Montal‐Mueller method, and the pore‐forming proteins activity is detected by polymeric electrodes. This methodology could be applied to examine different membrane proteins for the purpose of biosensing, drug screening and nanopore technologies.  相似文献

The excellent detection sensitivity in various matrices of T‐2 toxin (T‐2), which has cytotoxic and immunosuppressive effects in DNA and RNA synthesis, is a highly desirable characteristic. A sensitive molecularly imprinted electrochemical sensor was constructed for the selective detection of T‐2. In this study, iron ions (Fe3+) were introduced to increase the chelation of the metal ions and templates for preparing molecularly imprinted polymers (MIPs). With the increased chelation of the metal ions and templates, the selectivity and sensitivity of the MIPs were effectively improved. The imprinted sensor was successfully employed to detect T‐2 in cereals and human serum samples.  相似文献

Ts1 toxin is a protein found in the venom of the Brazilian scorpion Tityus serrulatus. Ts1 binds to the domain II voltage sensor in the voltage‐gated sodium channel Nav and modifies its voltage dependence. In the work reported here, we established an efficient total chemical synthesis of the Ts1 protein using modern chemical ligation methods and demonstrated that it was fully active in modifying the voltage dependence of the rat skeletal muscle voltage‐gated sodium channel rNav1.4 expressed in oocytes. Total synthesis combined with click chemistry was used to label the Ts1 protein molecule with the fluorescent dyes Alexa‐Fluor 488 and Bodipy. Dye‐labeled Ts1 proteins retained their optical properties and bound to and modified the voltage dependence of the sodium channel Nav. Because of the highly specific binding of Ts1 toxin to Nav, successful chemical synthesis and labeling of Ts1 toxin provides an important tool for biophysical studies, histochemical studies, and opto‐pharmacological studies of the Nav protein.  相似文献

取水产品样品2.00g与2g无水硫酸钠混匀后加入提取剂10 mL甲醇,涡旋振荡5min后超声提取5 min,再离心5 min,取其上清液。在上清液中加入500 mg中性氧化铝和15mg石墨化碳黑(GCB)涡旋振荡1min,离心5min,取上清液,于40℃下吹氮至近干,用体积比为1∶1的流动相A-流动相B混合溶液溶解残渣并定容至2.0mL,经0.22μm滤膜过滤,取其滤液按高效液相色谱-高分辨质谱法测定其中3种蓝藻毒素的含量。色谱分离中用Hypersile Gold C8色谱柱(150mm×2.1mm,3μm)为固定相,用不同比例的流动相A和流动相B两溶液的混合液作为流动相进行程序梯度洗脱。质谱测定中采用电喷雾离子源,正、负离子切换模式。由于3种蓝藻毒素均产生基质减弱效应,制作工作曲线需用基质标准溶液以消除基质效应。结果表明:所测3种蓝藻毒素均在10~200μg·L^-1内与其峰面积之间呈线性关系,其检出限(3S/N)均为5μg·kg^-1。通过标准加入法进行回收试验,测得回收率为68.3%~104%,测定值的相对标准偏差(n=6)为7.7%~17%。  相似文献
