CLONING, EXPRESSION AND CHARACTERIZATION OF Pro3 GENE OF YEAST Saccharomyces cerevisiae

The proline biosynthetic pathway and Pro genes in Saccharomyces cerevisiae have just begun to be studied recently. In our laboratory, Pro2 gene of S. cerevisiae had been cloned in yeast. As described in this paper, yeast Pro3 gene was also cloned, which can complement yeast Pro3 mutants, and be expressed efficiently in E. coli. The high activities of this gene product, L-pyrroline-5-carboxylate (P5C) reductase, can be detected in both organisms. The activity of the Pro3 gene product in multiple copy plasmids is not higher than that of single copy genes in chromosomes in both yeast and E. coll. The preliminary characterization of the gene is also reported.     

Improved staining of phosphoproteins with high sensitivity in polyacrylamide gels using Stains‐All

An improved Stains‐All (ISA) staining method for phosphoproteins in SDS‐PAGE was described. Down to 0.5–1 ng phosphoproteins (α‐casein, β‐casein, or phosvitin) can be successfully selectively detected by ISA stain, which is approximately 120‐fold higher than that of original Stains‐All stain, but is similar to that of commonly used Pro‐Q Diamond stain. Furthermore, unlike the original Stains‐All protocol that was time consuming and light unstable, ISA stain could be completed within 60 min without resorting to protect the gels from light during the whole staining procedure. According to the results, it is concluded that ISA stain is a rapid, sensitive, specific, and economic staining method for a broad application to the research of phosphoproteins.

非线性编辑方式与系统的广泛应用,极大地影响和改变了传统的电视节目制作方式,高度的系统兼容性、集成性与灵活性,加上丰富、简捷、高效的制作方法,为完美体现节目创作者的创作意图插上了腾飞的翅膀.由于计算机的视频信号处理能力与电视机并不完全一致,故应对非线性编辑制作的节目信号进行安全监控,使之符合传输与还原的要求.     

A new fluorescent quenching method for the determination of phosphoproteins by using calconcarboxylic acid

A fluorescent quenching detection method for phosphoproteins in SDS‐PAGE by using calconcarboxylic acid (CCA) was described. In this method, the fluorescence intensity of CCA was greatly increased with the presence of Al³⁺ in the gel background, while in zones where phosphoproteins are located this intensity was absent because of fluorescence quenching phenomenon through the formation of CCA‐Al³⁺‐phosphoprotein appended complex. Approximately 4–8 ng of phosphoproteins can be selectively detected within 1 h (1D SDS‐PAGE), which is similar to that of the most commonly used Pro‐Q Diamond stain. The specificity of this novel technique for phosphoproteins was confirmed by dephosphorylation, Western blot, and LC‐MS/MS analysis, respectively. Furthermore, to better understand the newly developed method, the detection mechanism of CCA stain was explored by fluorescent spectrometry. According to the results, it is believed that CCA stain may provide a new choice for selective, economical, MS compatible, and convenient visualization of gel‐separated phosphoproteins.     

基于OpenGL的水下云台可视化实现

用于控制声纳姿态的水下云台在实际应用中需要实现可视化。将设计云台的Pro/E模型保存为STL标准三维图形文件格式,利用3D Explore软件将该STL文件转换为标准数组。通过Visual C++结合Open GL编程技术,实现了水下云台的可视化,对准确控制云台提供了帮助。     

基于VLSI平台的MIPS处理器仿真与设计

传统ISA处理器由于其内部有限的逻辑资源和外部固定的引脚封装,大大地限制了它的应用范围.使用硬件描述语言,自底向上设计处理核心MIPS,并且与几类通用外设互连组成系统,使用Virtex?Ⅱ Pro系列FPGA进行板级验证.板级验证结果表明实现了既定目标,与标准MCU兼容,系统运行稳定.这样可以依据自己的需求,定制系统以达到传统MCU所无法完成的要求.
